Big Picture: Mitosis Definition and Function

  • Definition: Narrowly defined, mitosis is the part of eukaryotic cell division in which chromosomes within the nucleus are divided and transmitted to daughter cells. However, the term “mitosis” is frequently used as a synonym for “eukaryotic cell division” (and I do exactly that in this tutorial!). As an AP Bio student, you should be aware of both uses.
  • Functions of mitosis (broadly defined):
    1. Enables growth and development in multicellular organisms.
    2. Facilitates tissue repair and replacement.
    3. Allows reproduction in unicellular eukaryotes (e.g., amoebas).

The Cell Cycle

  • Divided into two main phases:
    1. Interphase (Preparation for division):
      • G1 Phase: Growth phase 1; the cell increases in size.
      • S Phase: Synthesis phase; DNA replication occurs.
      • G2 Phase: Growth phase 2; organelles and structures required for cell division are prepared.
    2. M Phase (Division):
      • Includes mitosis (chromosome separation) and cytokinesis (cytoplasm division).

A few key confusing terms 

  • Chromatin: The relaxed form of DNA during interphase.

    1 labels one half of an X-shaped molecule. 2 labels a horizontal band around 1 where the X crosses. 3 labels a series of horizontal lines that attach to 3. 4 labels a vertical red oval that connects both halves of 1.
    1. Sister chromatid. 2. Kinetochore. 3. Spindle fiber. 4. Centromere.
  • Sister Chromatids: Identical copies of a chromosome (before it’s pulled apart during mitosis). Sister chromatids are attached at a centromere.
  • Kinetochore: A protein structure on sister chromatids where spindle fibers attach.
  • Centrosome and Centrioles:
    • Centrosome: Also known as the microtubule organizing center, the centrosome creates microtubules, which are part of the cell’s cytoskeleton.
    • Within the centrosome are two centrioles, which (among their other functions) create a spindle of microtubules that separate the sister chromatids during mitosis.
    • Note from Mr. W: this is a very subtle difference. Being able to distinguish between centrioles and centrosomes is difficult for most AP Bio teachers, and probably not something students will need to do for the AP Bio exam.

Phases of Mitosis

  1. Interphase:
    • The cell grows and duplicates its DNA/chromosomes. Seen under a microscope, cells in interphase do not appear to be dividing.
  2. Prophase:
    • Chromatin condenses into visible chromosomes, consisting of two sister chromatids.
    • Nuclear membrane disintegrates.
    • Nucleolus (the site of ribosome assembly) disappears
    • Spindle apparatus begins to form from the centrosomes.
  3. Metaphase:
    • Spindle fibers attach to chromosomes at their centromeres.
    • Chromosomes are aligned at the cell’s equator (metaphase plate).
  4. Anaphase:
    • Spindle fibers pull the sister chromatids apart, moving them to opposite poles.
    • Non-kinetochore microtubules elongate the cell.
  5. Telophase:
    • New nuclear membranes form around each set of chromosomes.
    • Chromosomes relax into their interphase form (chromatin), and the nucleolus reappears.
  6. Cytokinesis:
    • The cytoplasm divides, resulting in two genetically identical daughter cells.
      • In animal cells a ring of protein fibers called microfilaments pulls together the membrane between the two daughter nuclei, splitting the cell in half
      • In plant calls a new cell wall grows between the daughter nuclei.

The Importance of the G0 Phase

  • G0 Phase: A resting state where cells exit the cell cycle and stop dividing.
    • Example: Highly specialized cells like nerve and muscle cells enter G0.
    • Some cells in G0 can reenter the cell cycle in response to specific stimuli.
